Makena (Hydroxyprogesterone Caproate Injection) Information
The FDA’s Center for Drug Evaluation and Research has proposed withdrawing approval of Makena, a hydroxyprogesterone caproate injection originally granted accelerated approval in 2011 to reduce preterm birth risk. A confirmatory trial—four times larger than the pivotal study—failed to demonstrate any benefit to newborn health or a reduction in preterm deliveries. The Obstetrics, Reproductive and Urologic Drugs Advisory Committee convened a virtual hearing on October 17‑19, 2022 to evaluate the data and decide whether the drug should remain on the market while further studies are designed. The FDA Commissioner will make the final determination based on the committee’s recommendations.
